Physiological response and transcriptome analysis of Prunus mume to early salt stress

Abstract Soil salinisation is an important factor limiting the cultivation and distribution of Prunus mume . Therefore, identifying regulation mechanism salt tolerance P. will promote its molecular breeding. In this study, garden tree species ‘Meiren’ was investigated, with a soil NaCl content (w/w) 0.3%. Photosynthetic gas exchange parameters, relative electrical conductivity, malondialdehyde, osmoregulation substances, antioxidant enzyme activity were measured in early (3d), middle (10d), late (30d) stages stress. Salt treatment applied for 0 h, 6 24 72 h; following this, gene library constructed from leaves. The results showed that CBL-CIPK, mitogen-activated protein kinase, calcium-dependent abscisic acid signalling pathways involved stress signal transduction Furthermore, bHLH, WRPK, ERF, MYB identified as potential key transcription families Combined determined physiological responses, inhibited activities RuBisCO, chlorophyll synthase, glutamyl tRNA reductase, divinyl magnesium chelatase; resultantly, led to reduction photosynthetic rate. Also, it found P5CS, SS, LEA, dehydrin regulated synthesis proline, soluble sugar, macromolecular alleviate osmotic POD plays role scavenging reactive oxygen due activity. findings research reveal internal response provide foundation construction genetic regulatory network
